With a segment triggered flow, when you set this flow live, the flow does not automatically backfill recipients in the segment and will only automatically trigger for new profiles who enter the flow after the flow is set Live.
In order to market to these profiles, please "add past profiles" in the Manage Flow section in the top right corner here:
You can find the prompt above by clicking the “Add Past Profiles” icon on the flow’s trigger node.
